再障患者血清和血宝胶囊对骨髓基质细胞-设计

【摘要】 目的:探讨血宝含药血清和再生障碍性贫血患者血清对骨髓基质细胞的影响。方法:提纯的再障患者血清抑制蛋白组份加到骨髓基质细胞培养体系48h后,采用免疫组化法观察骨髓基质细胞形态学观察和TUNEL法测定骨髓基质细胞的凋亡。大鼠胃饲血宝7天后制成含药血清按比例加入培养液,培养骨髓基质细胞,观察细胞形态和通过计数了解血宝对骨髓基质细胞的刺激作用;结果: 20例正常人骨髓基质细胞分别与0.1ug/ml,1ug/ml,10ug/ml的再障血清抑制蛋白组份共同培养后,Fas表达阳性率分别为(10.66±4.2)%(P>0.05),(16.92±9.8)%(P>0.05),(49.4±10.5)%(P<0.01),而对照组为(14.4±7.8)%;凋亡细胞发生率分别为(5.6±1.6)%(P>0.05),(24.82±8.2)%(P<0.01),(43.3±9.8)%(P<0.01),对照组为(4.6±1.5)% 。含药血清高,中,培养骨髓基质细胞5天后细胞计数分别(5±0.5)x104,(P<0.01),(2.3±0.6)X104(P<0.05),空白对照组为(1.3±0.2)X104 结论:再障患者血清抑制蛋白组份与正常人骨髓基质细胞的凋亡有一定的量效关系。推测Fas-FasL系统可能是启动骨髓基质细胞凋亡的一条重要途径,血宝具有促进骨髓基质细胞增殖作用。

Influence of Xuebao capsule and serum in aplastic anemia on bone marrow mesenchymal cell

【Abstract】 objective: To investigate Xuebao capsule and serum inhibitor in aplastic anemia influence on normal bone marrow mesenchymal cell Methods: Purified serum inhibitor in aplastic anemia was added into culture system of 7 days cultured normal bone marrow mesenchymal cell for 48 hours. Fas expression of normal bone marrow mesenchymal cell was assayed by immunohistochemistry, morphological change was observed and TUNEL was used to determine cell apoptosis .Mouse was feed with different dose “Xuebao”for five days and collected serum for cultivation bone marrow mesenchymal cell Results: Following culture of normal bone marrow mesenchymal cell in 20 cases combined with serum inhibitor of aplastic anemia 0.1μg/ml, 1μg/ml, 10μg/ml,we found all Fas expression positive rate was respectively(10.66±4.2)%(p>0.05),(16.92±9.8)%(P>0.05),(49.4±10.5)%(P<0.01),while the rate of the control group was(14.4±7.8)%;all apoptosis rate was respectively(5.6±1.6)%(P>0.05),(24.82±8.2)%(P<0.01),(43.3±9.8)%(P<0.01),while the rate of the control group was (4.6±1.5)%.
