【摘 要】目的:探讨独活寄生汤含药血清对大鼠退变软骨细胞Bcl-2、Bax表达的影响。策略:建立体外退变软骨细胞模型,分别给予独活寄生汤含药血清(含药血清组)和空白血清(空白血清组)干预24 h、48 h、72 h,收集软骨细胞,运用RT-PCR法检测血清干预后退变软骨细胞Bcl-2、Bax mRNA表达,Western Blot法检测血清干预后退变软骨细胞Bcl-2、Bax蛋白表达。结果:独活寄生汤含药血清对Bcl-2和Bax蛋白表达的影响与其对Bcl-2和Bax mRNA的影响具有相同的趋势,随着干预时间的延长,空白血清组和含药血清组Bcl-2蛋白阳性表达率均逐渐升高,含药血清组升高更显著;Bax蛋白阳性表达率逐渐降低,含药血清组降低更明显,且各时点之间比较,差异有统计学作用(P < 0.01)。结论:独活寄生汤能通过上调大鼠骨关节炎软骨细胞抑制凋亡因子Bcl-2表达,下调促凋亡因子Bax表达,抑制线粒体凋亡通路的激活,从而有效抑制软骨细胞凋亡。

Effects of Serum Medicated with Duhuo Jisheng Decoction on the Expression of the Degenerated Cartilage Cells Bcl-2 and Bax in Rats

【ABSTRACT】Objective:To observe the effects of serum medicated with Duhuo Jisheng Decoction on the expression of the degenerated cartilage cells Bcl-2 and Bax in rats.Methods:The degenerated cartilage cell models in vitro were established,which were intervened by the serum medicated with Duhuo Jisheng Decoction(the medicated serum group)and the blank serum (the blank serum group)for 24 h,48 h and 72 h,collecting chondrocytes.The RT-PCR method was used to detect the expression of degenerated cartilage cells Bcl-2 and Bax mRNA after serum intervention.The Western Blot method was used to detect the expression of degenerated cartilage cells Bcl-2 and Bax protein after serum intervention.Results:The effects of the serum medicated with Duhuo Jisheng Decoction on the expression of Bcl-2 and Bax protein had the same tendency as on the expression of Bcl-2 and Bax mRNA,.along with the extension of time,the positive expression rate of Bcl-2 protein in the two groups gradually increased,the medicated serum group being more significant than the blank serum group;while the positive expression rate of Bax protein in the two groups gradually decreased,the medicated serum group being more significant.The difference between each time point was statistically significant(P < 0.01).Conclusion:Duhuo Jisheng Decoction can inhibit the activation of the mitochondrial apoptosis pathway by up-regulating the expression of rat osteoarthritis cartilage cells Bcl-2 and down-regulating the expression of Bax,thus effectively inhibiting chondrocyte apoptosis.

骨关节炎(osteoarthritis,OA)又称退行性关节炎、增生性关节炎、肥大性关节炎等,是老年常见的骨关节疾病。随着社会老龄化的加剧,OA患者逐渐增加,已严重影响到人们的健康和生活。目前,普遍认为OA是在力学和生物学等多种因素共同作用下导致软骨细胞、软骨细胞外基质、软骨下骨三者降解与合成耦联失衡的结果[1],其中软骨细胞破坏及软骨基质降解是其主要病理过程。换言之,延缓软骨细胞及软骨基质降解是OA的主要治疗目的和策略之一。独活寄生汤出自《备急千金要方》,为治疗久痹而致肝肾两虚、气血不足证之验方,临床应用日益广泛,并获得良好疗效[2],但其确切作用机制尚未完全明了。本实验通过独活寄生汤含药血清对大鼠退变软骨细胞Bcl-2、Bax影响的研究,探讨本方延缓OA软骨退变的可能机制,为临床应用提供实验依据。 全文地址:www.7ctime.com/ywgllw/lw48277.html上一论文:阐述突发性耳聋应用鼻内窥镜下经咽鼓管置管注药治疗的疗效观察
